李国云,博士,教授,博士生导师。2009年毕业于中国海洋大学食品科学与工程专业,获得学士学位;2015年于中国海洋大学获得博士学位。2012至2014年在美国伦斯勒理工学院Robert J. Linhardt实验室开展糖分析化学方面研究。2015年7月通过中国海洋大学“青年英才工程”计划加入医药学院糖化学与糖药物研究团队。目前主要聚焦于复杂糖链的精细结构与功能研究,通过发展糖链分析方法学,解决糖类药物研发的关键技术问题,致力于海洋糖类创新药物研发。近年来在Anal. Chem., Carbohyd Polym, J Proteome Res等国际学术期刊发表SCI论文80余篇,授权国家发明专利14项(实现成果转让2项)。先后主持国家自然科学基金面上项目、青年项目、山东省自然科学基金等科研项目12项,参与国家重大新药创制、国家自然科学基金重大项目等国家级重大项目4项。 | ||||

糖组学是继基因组学和蛋白质组学后的新兴研究领域,是研究生物体内糖链结构与生物学作用的学科。糖链作为生命活动的重要信息分子,参与了真核生物几乎所有的生命过程。以糖为基础的新药开发已经成为本世纪国际生化药物研究的重点和热点。海洋糖类化合物具有资源丰富、结构特殊、能模拟细胞膜糖连且几乎没有毒副作用等特点,是新药开发的良好模板分子。本课题组以海洋生物为研究对象,运用现代色谱技术分离和纯化其中天然糖类化合物,并利用高分辨NMR和生物质谱结合糖生物信息学建立分析技术体系用于解析复杂多糖的结构,并通过化学、酶法等降解策略高效获得系列结构明确的海洋糖类药源分子,基于计算机模拟、糖芯片技术以及及细胞水平、动物水平等多效筛选及评价研究海洋糖类化合物在抗肿瘤、抗病毒、抗代谢性疾病等方面的构效关系和作用机制,阐明糖类药物体内代谢规律,为结构明确、功效明晰的新型海洋糖类药物创制,奠定坚实的理论基础。 糖组学研究与开拓已成为后基因组时代全面揭示生命本质的必由之路。但是由于生物样品中糖链含量甚微及其结构复杂性和特殊性,糖链精细结构分析技术一直是制约糖组学发展的瓶颈。本课题组利用现代色谱分离技术和生物质谱技术及糖生物信息学等糖组分析研究前沿技术,建立微量生物样品中糖链(包括糖脂、N-糖链、O-糖链和GAGs)高效、超灵敏定量分析体系,并在细胞水平、动物模型和临床样本多个层次筛选和鉴定肿瘤的特征性糖链,研究相关糖链在肿瘤中的表达、结构特征、功能特点及其变化规律,为肿瘤诊断和治疗的研究提供新策略和新靶点。 | ||||
